Yes, but it requires some work:

NOTE: In order for this to work, you must first go Control Panel, click Appearance and Personalization, then click Folder Options, click the View tab, and make sure the check box "Hide extensions for known file types" is unchecked. (I'm using Windows Vista for this by the way).

Step 1 – Open iTunes, right click the song you want as a ringtone, and select Get Info.

Step 2 – Click on Options and select a start and end time to a no more than 40 second margin.

Step 3 – Click OK, then close the window, right click the song again and click "Convert song to AAC."

Step 4 – Right click on the newly converted file and select "Show in Explorer" (or Finder if on a MAC), keep the newly opened window open, but then go back to iTunes and delete the converted file from iTunes, but when prompted, select "Keep file."

Step 5 – Go to the file in Explorer or Finder and change the file name ending to m4r, then when prompted, select "Keep file name."

Step 6 – Double click on the song in Explorer or Finder and it will appear in the Ringtone section of iTunes.

    I'm not certain if there's a program for Windows…

    Jailbreak or get a Mac =P.

    Jailbreak has a program that allows you to make ringtones.

    Macs come with Garageband which lets you make ringtones for free (provided they aren't DRM encrypted)

    Just select your phone as an iPhone
    They will email you the ringtone
    FREE!!!
    Once they email you the ringtone, open the attachment, it will open it up in iTunes, and just sync your iPhone to your computer!!
    They will let you create your ringtone out of mp3 files (Files in iTunes,etc.)

    To use this website, YOU DON'T HAVE TO HACK/JAILBREAK YOUR IPHONE.
